Antidepressant Nonresponders
Completing Trial of Buspirone Potentiation
- The next two slides
present results from an open series which investigated the ability of
a medium buspirone dose (30 mg/d) to potentiate antidepressant response
in nonobsessional affective disorder patients who were refractory to
standard trials of antidepressants (DSM-IIIR).
- Eight patients
were started on buspirone (30 mg/d); dosage was reduced if side effects
were intolerable.
- Seven patients
were treated with the SSRI fluoxetine and one patient was treated
with the TCA imipramine.
- Patients were
evaluated within 3 weeks of treatment initiation.
- A good response
was defined as a remission of depressive symptoms, as reflected by a
reduction in Clinical Global Impression (CGI) scores (which measure
severity of depression), by 3 or more points to 1 (not ill).
- Partial response
was reflected by a 2-point reduction in CGI score.
- This slide shows
that four patients (50%) achieved complete resolution of depressive
symptoms (CGI=1); three patients (37%) experienced partial response;
and one patient (13%) demonstrated no response.
- The posttrial
group mean CGI score was 1.9 (1.4, P 0.001).
- Reported side
effects were negligible.
